"Biological Light Microscopy in 2020: Counting photons"
Abstract
All microscopical techniques are characterized by two processes: localization and measurement.
Recently, a number of new techniques have allowed us to break the Abbe-diffraction barrier and increase the precision of localization. Less attention has been paid to measurement, a process whose accuracy is limited, on the one hand, by Poisson statistics and on the other hand by bleaching/phototoxicity.
This talk will highlight the limitations imposed on live-cell microscopy by the need to measure light intensity, so we can consider the best way to proceed.
