Reflection-Contrast Microscopy for high definition images with light microscopy: Practical tips using new Leica DMR equipment for RCM

he purpose of this paper has been to illustrate with some examples of practical possibilities and difficulties, how a user could choose an optimal RCM-setting for his applications. Further adjustments of the RC microscope set-up, are then rarely needed. RCM is then a practical proposition for routine use in for example a pathology laboratory.
In a number of applications we came to the conclusion that RCM provides a significant increase in image definition, compared with most conventional light microscopical methods. This allowed often the observation of subcellular structures with RCM that up till now were only identified with confidence using TEM.
