Based on extensive customer feedback, Leica Microsystems has created a new device called the Leica Cryosphere which minimizes contamination of sections by ice crystals during cryosectioning with the Leica EM UC6 Ultramicrotome/Leica EM FC6 Cryo Chamber.
A transparent sealed structure covers the cryo-ultramicrotome. The nitrogen gas produced for cooling the EM FC6 chryochamber, specimen and knife are sufficient enough to deplete the moisture in the cryochamber to <1%.
Good quality biological cryosections require a humidity of less than 20%, but preferably zero.
