In this workshop, we will introduce the new THUNDER Imager Cell Spinning Disk, which allows high-speed, high-resolution imaging with reduced phototoxicity. While spinning disk alone provides fast, versatile, confocal imaging and powerful optical sectioning, its new synergistic pairing with THUNDER eliminates residual out-of-focus light, resulting in greater contrast and resolution. This enables you to extract more statistically significant data from diverse 3D samples in less time and with reduced phototoxicity. We will demonstrate this using a practical example of 3D human urothelium organoids. We will also introduce the intelligent automation features of the THUNDER Imager Cell Spinning Disk, including Adaptive Immersion and SmartCORR. Finally, we will discuss how combining this with Aivia for AI-based image analysis can streamline workflows, improve image quality, and deliver more reproducible results. 

Precision sectioning redefined: bringing ultramicrotomy to the digital age

We will introduce the Leica UC Enuity ultramicrotome, a revolutionary advancement in electron microscopy sample preparation. Building on its predecessors' strengths, it features integration of automated setup functions, including auto-alignment of the block face and knife, and auto trimming. These significantly reduce the time and expertise required for sample preparation, making high-quality sectioning accessible to users of all skill levels. The UC Enuity’s modular design is fully upgradable, allowing it to evolve with the user's needs and technological advancements. A new is its ability to utilize micro-CT based 3D trimming, which precisely targets specific regions within a sample, using 3D micro-CT data to guide the trimming process. This innovative approach saves time, reduces sample loss and minimizes the need for extensive user training.

Join us to explore the experimental design and considerations for performing high multiplexing imaging experiments beyond 15+ fluorophores on a single sample on the STELLARIS platform. Topics include appropriate fluorophore panels, sample preparation, and the challenges and tools required for imaging. We will show how imaging high multiplexing targets on a single sample in one go (one round of labeling and imaging) can be exploited for 3D spatial omics analysis. Furthermore, we will discuss how AI-based image analysis using Aivia software enables robust and efficient analysis of high multiplexing imaging data, providing powerful tools to extract meaningful insights from complex spatial biology experiments.
