The Leica DM6 FS fixed stage microscope is designed for neuroscience applications such as electrophysiology. The system has special adaptations, such as convenient specimen access, flexibility in design, and electric stability. For example, it switches off all motors and current-carrying elements during data acquisition for the highest mechanical and electronic stability during your experiments.

Furthermore the Leica DM6 FS is an ideal instrument for any fluorescence imaging purpose where the specimen is in an aqueous solution and cannot be imaged using an inverted microscope. For these applications Leica Microsystems offers special high performance water dip-in lenses. Examples include thick or non-transparent specimens such as tissue slices, whole organs, or whole animals.

The Leica DM6 FS is highly automated. For example, a motorized objective changer allows vibration-free changing between two objectives. While switching magnification, you do not have to touch the instrument at all. An access angle of 41° to 45°and a free working distance of between 2.5 mm and 3.7 mm provide excellent clearance around the specimen.

In addition, the combination of a magnification changer with “low magnification – high numerical aperture objectives”, such as the 25x/0.95 NA water immersion objective, allows for optical switching between magnifications in transmitted light mode during an experiment without changing the objective at all.

The Leica DM6 FS is tailored for electrophysiology. It has special shielded cables and motors plus space for peripherals. You can combine Differential Interference Contrast (DIC) and epifluorescence with the recording or synchronization of electric signals. Your experiments remain stable, because the instrument eliminates all mechanical vibrations and electrical interference. You can easily access the specimen because of the large amount of space around the specimen. You can also switch objectives without touching the microscope, using the motorized two-position objective changer and gently submerge the objectives into the aqueous solution with precise automatic refocusing. Last, but not least, the Leica DM6 FS is ready for Optogenetics when combined with equipment such as the Lumen Dynamics X-Cite XLED1 or Andor Mosaic or Rapp OptoElectronic UGA-42-Firefly.

Show answer Are there special objectives available for electrophysiology?

Yes. Electrophysiological applications often need objectives with an extra-long free working distance. These are designated by an L in the objective name. Leica Microsystems offers a range of objectives with ample free working distance for dry or water immersion. The water immersion objectives with extra-long working distances also deliver a large access angle and an inert ceramic front with minimal electric and thermal conductivity for electrophysiology.

The U-V-I objectives have very good transmission in the whole wavelength range, especially in the (near) UV-range. This makes them perfectly suited for applications like Fura-2 calcium imaging.
