Tune into any Excitation with Supercontinuum Confocal Leica TCS SP5 X

The Leica TCS SP5 X ‘white light laser’ confocal can excite the dyes of today and tomorrow with ideal spectral detection.

With the Leica TCS SP5 X researchers can tune excitation wavelengths to perfectly match fluorophores, reduce laser power, and increase viability for long live cell experiments.

The tunability of excitation and detection, allows acquiring complete excitation emission spectra. With this spectral information, any dye can now be optimally excited with minimum cross-excitation and specimen damage. Up to eight excitation lines can be used simultaneously.

The pulsed White Light Laser (WLL) supplies also excitation light for FLIM. The fluorescence lifetime provides information about the direct environment of a molecule, like local pH or binding to another molecule.

The choice of fluorescent labels is not limited any longer by the hardware. The researcher selects the dyes according to their availability or the scientific needs. With up to five spectral detection channels, the Leica TCS SP5 X is completely tunable and fully adapts to the specimen.

New dyes or fluorescent proteins can be discovered and characterized. A two-dimensional excitation-emission spectrum characterizes the spectral properties of any fluorescent sample. Fluorophores with identical emission or excitation spectra are discriminated. This is essential for multi-color fluorescence experiments.

Optimal excitation and detection

Spectral optimization of excitation and detection in samples with multiple fluorescent species results in minimized cross excitation and crosstalk combined with maximum excitation and detection efficiency. This reduces acquisition time and photo bleaching. The artifact reduction enhances data quality and the information content of the images.

Free selection of FLIM excitation wavelength

The White Light Laser is an appropriate excitation source for fluorescence lifetime measurements. Its continuous tunability allows performing FLIM measurement at any excitation wavelength. Lifetime of orange and red dyes can now be obtained.
